自噬是一种由应激诱导的保护性机制。在基础条件下活性较低;当细胞面临营养耗竭、化学或环境胁迫等特定不利条件时,会启动该机制,通过溶酶体介导大量降解细胞内容物。自噬活性增强在癌症、神经退行性疾病、心血管疾病和糖尿病病理中的作用已得到广泛认可和普遍研究。
CYTO-ID® Autophagy detection kit利用一种新型染料,选择性标记积累的自噬泡,从而检测自噬泡并监测自噬流,适用于溶酶体功能受抑制的活细胞。这种可被488nm激发的绿色染料经过优化,通过鉴定其可调控的功能基团,实现了对溶酶体的极微染色,同时在结合前自噬体、自噬体和自噬溶酶体时呈现明亮的荧光。

PROTEOSTAT® Aggresome detection kit(货号:ENZ-51035)
Aggresome是由聚集的错误折叠蛋白质形成的包涵体,通常在泛素-蛋白酶体蛋白质降解机制失效时形成。Aggresome是细胞为应对应激而产生的一种细胞保护机制,通过隔离错误折叠的蛋白质,最终使其通过自噬途径被清除。此外,蛋白质Aggresome的形成也是多种人类疾病的标志,如阿尔茨海默病、帕金森病、肌萎缩侧索硬化症或酒精性肝病等。
PROTEOSTAT® Aggresome detection kit所含的PROTEOSTAT® 检测试剂是一种分子染料。 在溶液中,染料分子沿单一中心键的自由旋转会抑制荧光。PROTEOSTAT®染料能特异性嵌入到四级蛋白质结构特有的交叉β-折叠骨架中——这种结构常见于错误折叠和聚集的蛋白质——并产生强烈的荧光。该试剂盒提供了一种快速、特异且定量的方法来标记细胞内的Aggresome,无需对蛋白质进行人工突变。
