在单克隆抗体的制备过程中，杂交瘤细胞的筛选和鉴别是最重要的步骤。首先需要筛选出正确融合的杂交瘤细胞，再筛选出能分泌目标抗体的杂交瘤细胞，最终将筛选出来的杂交瘤细胞进行扩大培养，制备大量单克隆抗体。在进行能分泌目标抗体杂交瘤细胞的筛选时，需要鉴定这类杂交瘤细胞培养上清液中单克隆抗体的类或亚类，最-常-用的是ELISA法，因其快速、简便、灵敏度高、特异性强、易于标准化等优点，已得到广泛应用。
